IMO, NYC isn't as much of a "must see" kind of place as it is a "must experience" kind of place - the must experience being the various neighborhoods and what they have to offer (East Village, West Village, Union Square, SoHo, Flatiron, Meat Packing District, Gramercy, Tribecca, Midtown, Upper East Side, Lower East Side and Upper West Side). Some of the best restaurants in the world are here; Danielle, Per Se, Gramercy Tavern, Le Bernadin, Nobu, Morimoto, Craft... the list goes on. Also, the nightlife is great depending on what you're into. You can do the "models and bottles" scene, right now some good spots for that are Greenhouse on Tuesdays, The Box on Saturdays, The Garden at Ono (downstairs in the Gansevoort Hotel) on Saturdays and High Bar after 5PM on the weekdays. For a more scenesterish experience you can hang out in the East Village and Lower East Side. Some great restaurants in the East Village are Yuca Bar (Latin / Cubin), Franks (Italian), Momofuku (noodles), Tasho (yakitori), and Pauls (burgers).
I'm gonna stop here because I realize I could write pages of recommendations, but if you can narrow down what types of places you're into maybe I can be a little more helpful, lol.
